Background and Aims: Aging is associated with metabolic alterations and impaired pancreatic beta cell function. Pancreatic and duodenal homeobox 1(PDX1) plays an important role in beta cell differentiation, survival, and function, and alterations in its expression may accompany pancreatic dysfunction. This study aimed to investigate the effect of epigallocatechin-3-gallate (EGCG) on PDX1 gene expression in the pancreatic tissue of adult male Wistar rats subjected to a D-galactose-induced aging model.
Materials and Methods: This experimental study included 40 adult male Wistar rats that were randomly allocated to four groups, namely control, EGCG, D-galactose, and D-galactose + EGCG. Aging was induced by intraperitoneal administration of D-galactose at 100 mg/kg body weight for 9 weeks. In the D-galactose + EGCG group, EGCG was administered after induction of the aging model at 25 mg/kg body weight by oral gavage for 4 weeks. Following treatment, RNA was extracted from pancreatic tissue, and cDNA was synthesized. PDX1 expression was evaluated using real-time PCR and the 2−ΔΔCt method, with GAPDH as the reference gene. Data were analyzed using one-way analysis of variance followed by Tukey’s post hoc test, with statistical significance set at P<0.05.
Results: Relative PDX1 expression decreased in the D-galactose group, compared to the other groups. Moreover, EGCG administration after induction of aging increased PDX1 expression in the D-galactose + EGCG group, compared to the D-galactose group (P=0.036). The RQ values were 1.01, 1.67, 0.35, and 0.69 in the control, EGCG, D-galactose, and D-galactose + EGCG groups, respectively.
Conclusion: Following induction of aging, EGCG administration was associated with increased PDX1 expression in rat pancreatic tissue. Given the established role of PDX1 in maintaining β-cell identity and function, this finding may indicate that EGCG modulates molecular pathways involved in pancreatic β-cell homeostasis. Assessment of PDX1 protein levels and pancreatic functional indices may further clarify the biological significance of these molecular changes.
