Background: In recent years, the incidence of liver damage due to improper diet, and sometimes the use of drugs and hepatotoxic substances has been increased. Considering that so far, limited studies have been evaluated protective interventions against the metabolic side effects of glucocorticoids, especially Non-Alcoholic Fatty Liver Disease (NAFLD), and crocin might also be beneficial in preventing liver damage due to consuming a high-fat diet, in this study the effect of crocin on lipid metabolism of mice treated with glucocorticoids was investigated. Methods: This study was conducted on 30 eight-week-old male mice weighing (25-30 grams) in 5 equal groups. The study groups included the control, High-Fat Diet (HFD), glucocorticoids (GC) + HFD, GC + HFD + crocin (50 mg/kg), and GC + HFD + crocin (100 mg/kg). The total duration of the study was 13 weeks, and the last two groups received crocin intraperitoneally from the 8th to the 13th week each day. At the end of the 13th week, after a night of fasting, the animals were sacrificed with pentobarbital (60 mg/kg). Blood sample was taken to measure the levels of triglyceride and total cholesterol. Pre-renal and gonadal fat tissue was also examined for histological studies. Results: Based on the results, the HFD consumption and corticostron significantly increased cell size of pre-renal and gonadal fat pads. Crocin could significantly reduced these variations in pre-renal fat dose dependently (p<0.0001). HFD also significantly increased lipid index, serum cholesterol and triglyceride ((p<0.001) levels. Crocin not improved serum cholesterol level and fat index however, it controls high serum triglyceride level. Conclusions: based on present study, crocin was able reduced the fat cells size and improved lipid metabolism although had no beneficial effect on serum cholesterol level.
