fa اثر پروتئین NS3 ویروس هپاتیت سی بر القای بیان نشانگرهای HSP70 و Glypican3 کارسینومای هپاتوسلولار Effects of hepatitis C virus NS3 protein on expression of heat shock protein 70 and Glypican3 as the markers of hepatocellular carcinoma ويروس شناسي Virology مقاله اصیل پژوهشی Original Article <div style="text-align: justify;">زمینه و هدف: عفونت ویروس هپاتیت C (HCV) یک عامل خطر مهم در ایجاد سرطان کبد است. پروتئین غیرساختاری 3 (NS3) ویروس HCV که نقشی کلیدی در چرخه زندگی ویروس ایفا می‌کند، می‌تواند بر فعالیت‌های طبیعی سلولی مانند: تکثیر سلول، مرگ سلولی و مسیرهای پیام رسانی سلولی تأثیر بگذارد همچنین شاید بتواند در ایجاد بدخیمی مؤثر باشد. از سوی دیگر دو ژن سلولی Heat Shock Protein 70 (HSP70) و Glypican3 (GPC3) که در این مطالعه ارزیابی می‌شوند، نقش‌های مهمی در تنظیم مسیرهای پیام‌رسانی سلولی از جمله تکثیر سلولی دارند. هدف از این مطالعه، ارزیابی تأثیر پروتئین HCV NS3 بر بیان این دو ژن در مدل موش <a name="_Hlk87294094">Balb / C</a> بود. روش تحقیق: در این مطالعه، سه گروه موش نر  Balb / C (n=8) در نظر گرفته شد: گروه اول که پلاسمید NS3 را دریافت کردند، گروه دوم که پلاسمید HBx ویروس هپاتیت B را دریافت کردند و گروه کنترل منفی که آب مقطر دریافت نمودند. دو تزریق از طریق ورید دم انجام شد و پس از آخرین تزریق، RNA از بافت کبد استخراج شد، سپس سنتز cDNA و Real Time PCR برای ژن‌های مربوطه صورت گرفت. یافته‌ها: نتایج نشان داد که بیان نسبی ژن‌های انتخاب شده در گروه NS3 در مقایسه با گروه کنترل منفی معنی‌دار بود. (برای ژن GPC3، 0/0229=P و برای ژن HSP70، 0/0020=P) امّا تفاوت معنی‌داری بین گروه NS3 در مقایسه با گروه HBx وجود نداشت (برای ژن GPC3، 0/4516=P و برای ژن HSP70، 0/6740=P).   نتیجه‌گیری: نتایج به‌دست آمده نشان داد که NS3 شاید بر افزایش بیان ژن‌های ذکر شده مؤثر باشد؛ امّا برای فهم دقیق‌تر، مطالعات بسیار بیشتری باید انجام شود؛ مانند: ارزیابی تأثیر NS3 بر سایر پروتئین‌های درگیر در مسیرهای پیام‌رسانی سلولی، مطالعه سایر دومین‌های NS3، انجام آزمایش‌های پاتولوژیک و بافت شناسی، تغییر روش‌های آزمایش، ارزیابی نقش NS4A به عنوان یک فاکتور برای NS3 و استفاده از وکتورهایی با پایداری بیشتر.</div> <div style="text-align: justify;">Background and Aims: Hepatitis C virus (HCV) infection is an important risk factor for the development of liver cancer. The HCV NS3 protein plays a key role in the virus life cycle and can affect normal cellular activities, such as cell proliferation, cell death, and cell signaling pathways. Moreover, it may influence malignancy development. Two cellular genes, heat shock protein 70 (HSP70) and Glypican3 (GPC3), that are assessed in this study, play important roles in the regulation of the cell signaling pathways, including cell proliferation. This study aimed to evaluate the effects of HCV NS3 protein on the expressions of these two genes in the Balb/C mouse model. Materials and Methods: This study was performed on three groups of male mice of Balb/C (n=8). The first group received NS3 plasmid, the second group received hepatitis B virus HBx plasmid, and the negative control group received distilled water. Two injections were administered via the tail vein, and after the last injection, RNA was extracted from the liver tissue. Next, the cDNA synthesis and real-time polymerase chain reaction for relevant genes were performed. Results: Findings revealed that the relative expression of the selected genes in the NS3 group was significant in comparison with the negative control group (P=0.0229 for GPC3 and 0.0020 for HSP70). However, there was no significant difference between the NS3 group and the HBx group (P=0.4516 for GPC3 and 0.6740 for HSP70). Conclusion: Results showed that NS3 protein may affect the increasing expression of the mentioned genes.
