fa بررسی همزمان الگوی متیلاسیون ژن CADM1 و کمیت ویروس HPV به‌ عنوان بیومارکر تشخیصی در سرطان سرویکس Investigation of simultaneous of CADM1 gene methylation pattern and HPV virus quantification as a diagnostic biomarker in Cervical Cancer ژنتيك پزشكي Medical Genetics مقاله اصیل پژوهشی Original Article زمینه و هدف: پژوهش ها نشان داده اند که متیلاسیون پروموتر ژن CADM1، یکی از مکانیسم‌های اصلی خاموش ‌شدن بیان آن در سلول‌های سرطانی است. هدف از این مطالعه، تعیین الگوی متیلاسیون پروموتورژن CADM1 و تعیین کمیت ویروس پاپیلومای انسانی بود. روش تحقیق: در این مطالعه، 75 نمونه بافت توموری و 75 نمونه بافت حاشیه تومور که با تشخیص قطعی سرطان سرویکس از بیماران گرفته شده بودند، مورد مطالعه قرار گرفت. پس از استخراج DNA و اطمینان از کیفیت و کمیت DNA استخراج‌شده با روش اسپکتوفتومتری و الکتروفورز ژل آگارز، الگوی متیلاسیون CADM1 به روش HRM انجام گرفت.تعیین الگوی متیلاسیون ژن CADM1 بدون در نظر گرفتن نوع واریانت‌های درگیر با استفاده از روش Eva Green و مقایسه نقطه ذوب با درجه تفکیک بالا صورت گرفت و کمیت HPV پس از جداسازی آن از بلوک بررسی گردید. یافته‌ها: متیلاسیون ژن CADM1 در سلول‌های تومور سرویکس در مقایسه با حاشیه تومور، قبل و بعد از رادیوتراپی افزایش معنی‌داری داشت (P<۰/۰۰۰۱)؛ اما تغییر در میزان متیلاسیون سلول‌های توموری، قبل و بعد از رادیوتراپی معنی‌داری نبود. به این معنی که رادیوتراپی در میزان متیلاسیون ژن CADM1 تأثیری نداشت. همچنین میزان متیلاسیون ژن CADM1 در بیماران با تیتر HPV بالای ml/IU 500 به‌طور معنی‌داری بیشتر از بیماران با تیتر پایین‌تر از ml/IU 500 بود. نتیجه‌گیری: استفاده از DNA متیله سلول‌های سرطانی به‌ عنوان پیش‌آگهی برای رخداد سرطان سرویکس در زنان HPV مثبت مناسب است؛ همچنین می‌توان از ژن CADM1 نیز به‌عنوان یک نشانگر سرطان سرویکس استفاده نمود.   Background and Aim: Researches have shown that promoter methylation of the CADM1 gene is one of the main mechanisms is to silence its expression in cancer cells. The objective of this study was to determine the methylation pattern of the CADM1 gene promoter and to quantify the human papillomavirus. Materials and Methods: In this research, 75 tumor tissues and 75 tumor marginal tissue samples taken from patients with definitive diagnosis of cervical cancer were examined. After extraction of DNA and ensuring the quality and quantity of DNA extracted by spectrophotometry and the Agarose gel electrophoresis method, the methylation pattern of the CADM1was performed by the HRM method. Determining the methylation pattern of the CADM1 gene, without considering the type of involved variants was performed by using the Eva Green method and comparing the high-resolution melting point and HPV quantities were determined after isolation from the block. Results: Methylation of CADM1 gene in cervical tumor cells increased significantly (P<0.0001) in comparison with the tumor margin before and after radiotherapy. But the change in the level of methylation of tumor cells before and after radiotherapy was not significant. This means that radiotherapy does not affect the level of methylation of the CADM1 gene. Moreover, the level of methylation of CADM1 gene in patients with HPV titer higher than 500 ml/IU was significantly more than that of patients with HPV titer lower than 500 ml/IU.
