اثر ال-آرژینین بر نفوذپذیری اندوتلیوم آئورت و سطح سرمی لیپوپروتئینها در موشهای تغذیه شده با رژیم عادی و پر کلسترول
The effect of dietary L-Arginine on aortic endothelial permeability and level of blood lipoprotein in high-cholesterol fed rats

<p> <strong> زمینه و هدف: </strong>نیتریک اکساید، مادهای است که رگها را در برابر اتفاقات آتروژنیک محافظت میکند. مطالعات قبلی نشان داده است که رابطه مستقیمی بین افزایش نفوذپذیری سلولهای اندوتلیال و ایجاد و پیشرفت آترواسکلروز وجود دارد. مطالعه حاضر با هدف تعیین نقش ال- آرژینین بر نفوذپذیری سلولهای اندوتلیال آئورت و سطح سرمی لیپوپروتئینها در موشهای تغذیهشده با رژیم عادی و پرکلسترول انجام شد.</p>
<p> <strong> روش تحقیق: </strong>در این مطالعه تجربی، 36 سر موش نر به دو گروه آزمون و شاهد تقسیم شدند که هر گروه خود شامل دو زیرگروه دریافتکننده آب معمولی و آب به همراه 25/2% ال- آرژینین بودند. این رژیم غذایی به مدت هشت هفته ادامه داشت. در پایان هفته هشتم شریان کاروتید راست حیوانات کانوله شد و محلول ایوانبلو با دوز mg/kg 20 تزریق گردید؛ سپس تمام آئورت آنها برداشته شد و در 5/2 میلیلیتر محلول فرمامید قرار داده شد. پس از گذشت 24 ساعت، جذب نوری محلول فرمامید توسط اسپکتوفتومتر سنجیده شد. دادههای جمعآوری شده با استفاده از نرمافزار SPSS و آزمون یک طرفه ANOVA و نیز آزمون پس آماری Tukey در سطح معنیداری 05/0>P مورد تجزیه و تحلیل قرار گرفتند.</p>
<p> <strong> یافتهها: </strong>نتایج نشان داد که ال- آرژینین سبب کاهش نفوذپذیری اندوتلیوم آئورت در موشهای تغذیه شده با رژیم پرکلسترول در مقایسه با گروه شاهد شد (005/0>P)؛ همچنین در مقایسه با گروه شاهد مصرف ال- آرژینین موجب پایینآمدن سطح LDL در سرم خون موشهای تغذیهشده بارژیم پرکلسترول گردید (005/0>P).</p>
<p> <strong> نتیجهگیری: </strong>مصرف ال- آرژینین میتواند اثر حفاظتی بر نفوذپذیری اندوتلیوم عروق در حیوانات تغذیه شده با رژیم پرکلسترول داشته باشد.</p>
<p> <strong> Background and Aim: </strong>Nitric oxide (NO) is a substance which protects vessels against atherogenic effects. Previous studies have shown that there is a direct relationship between increasing of permeability of endothelial cells and formation and progression of atherosclerosis. The aim of this study was to evaluate the role of L-Arginine in permeability of aortic endothelial cells and serum lipoprotein level in high-cholesterol fed rats.</p>
<p> <strong> Materials and Methods: </strong>In this experimental study, 36 male rats were divided into two groups one with a normal and the other with a high-cholesterol diet .Then each group was divided into two sub-groups :Group “a” used L-Arginine 2.25% in water and group”b” used safe drinking water. After eight weeks, the right carotids of the animals were cannulated and 20 mg/kg of Evans Blue was injected into the subjects. Then, the whole aorta of each animal was dissected and put in 2.5ml formamid solution. After 24 hs, absorbance of the solution was measured by means of spectophotometer. Finally, the obtained data was analysed by SPSS software using one-way ANOVA, and Tukey statistical test and P≤0.05 was taken as the significant level.</p>
<p> <strong> Results: </strong>L-Arginine decreased endothelial permeability in aorta in rats feeding on high-cholesterol foods in comparison with the control group. (P<0.005). In addition, L-Arginine reduced the LDL level in the blood serum of rats with a high-cholesterol diet in comparison with the control group (P<0.005).</p>
<p> <strong> Conclusion: </strong>Administration of L-Arginine has a protective effect on the permeability of endothelium of aortic vessels in hypercholesterolemic condition.</p>
